چکیده

Axin antagonizes the developmental effects of Wnt in vertebrates. We show here that Axin simultaneously binds two components of the Wnt pathway, b-catenin and its negative regulator glycogen synthase kinase-3b. In mammalian cells, Axin inhibits Wnt-1 stimulation of b-cateniny lymphoid enhancer factor 1-dependent transcription. Axin also blocks b-catenin-mediated transcription in colon cancer cells that have a mutation in the adenomatous polyposis coli gene. These findings suggest that Axin, by forming a complex with b-catenin and glycogen synthase kinase-3b, can block signaling stimulated by Wnt or by adenomatous polyposis coli

A divergent canonical WNT-signaling pathway regulates microtubule dynamics

Dishevelled (DVL) is associated with axonal microtubules and regulates microtubule stability through the inhibition of the serine/threonine kinase, glycogen synthase kinase 3beta (GSK-3beta). In the canonical WNT pathway, the negative regulator Axin forms a complex with beta-catenin and GSK-3beta, resulting in beta-catenin degradation. Inhibition of GSK-3beta by DVL increases beta-catenin stabi...

b-Trcp couples b-catenin phosphorylation-degradation and regulates Xenopus axis formation

Regulation of b-catenin stability is essential for Wnt signal transduction during development and tumorigenesis. It is well known that serine-phosphorylation of b-catenin by the Axin–glycogen synthase kinase (GSK)–3b complex targets b-catenin for ubiquitination–degradation, and mutations at critical phosphoserine residues stabilize b-catenin and cause human cancers. How b-catenin phosphorylatio...

Convergence of cMyc and b-catenin on Tcf7l1 enables endoderm specification

The molecular machinery that directs formation of definitive endoderm from pluripotent stem cells is not well understood. Wnt/ b-catenin and Nodal signalling have been implicated, but the requirements for lineage specification remain incompletely defined. Here, we demonstrate a potent effect of inhibiting glycogen synthase kinase 3 (GSK3) on definitive endoderm production. We find that downstre...

Galangin suppresses the proliferation of β-catenin response transcription-positive cancer cells by promoting adenomatous polyposis coli/Axin/glycogen synthase kinase-3β-independent β-catenin degradation.

Galangin is a naturally occurring bioflavonoid with anticancer activity against certain human cancers, yet little is known about its mechanism of action. Here, we used a chemical biology approach to reveal that galangin suppresses β-catenin response transcription (CRT), which is aberrantly up-regulated in colorectal and liver cancers, by promoting the degradation of intracellular β-catenin. Inh...

A Role of Dishevelled in Relocating Axin to the Plasma Membrane during Wingless Signaling

Wnt signaling causes changes in gene transcription that are pivotal for normal and malignant development. A key effector of the canonical Wnt pathway is beta-catenin, or Drosophila Armadillo. In the absence of Wnt ligand, beta-catenin is phosphorylated by the Axin complex, which earmarks it for rapid degradation by the ubiquitin system. Axin acts as a scaffold in this complex, to assemble beta-...
